The Wikipedia article on Toyokuni II claims that one can distinguish his signature from that of Toyokuni I as the sides of the upper radical in 'Toyo' slope out (as does this one), unlike those of Toyokuni I, which are vertical.
I have no idea if this is true of not, I have yet to survey a large number of prints which can be securely attributed to each, but I thought I'd point it out, as if true, it would be an easy way to tell them apart.
