There is another impression of this sheet in the ARC database at Ritsumeikan University:

They assign the design to the play "Ura Omote Yanagi no Uchiwae" (裏表柳団画), performed at the Nakamuraza in August 1875. The casting looks right, but neither the role names nor costumes seem to line up with what I have found about that play, so I'm not sure what to think.

Anyway, here's what I see on the two sheets you've posted:

Nakamura Nakazō III 中村仲蔵 as 護待院
Ichikawa Danjurō 市川団十郎 as 伊井かもんの守 ("Ii kamon no kami"?)
Iwai Hanshirō VIII 岩井半四郎 as 操御前 ("Misao Gozen"?)
